Human PBMCs were cultured with 0.5× or 1× Cell Stimulation Cocktail (Protein Transport Inhibitors Plus, 500×) for 6 hours. Cultures were harvested, fixed and permeablized. The cells were then stained with APC Anti-Human IL-2 and analysed by flow cytometry.

细胞刺激混合物（含蛋白转运抑制剂，500×）包含佛波酯（PMA，PKC 激活剂）、离子霉素（Ionomycin）、布雷非德菌素 A（Brefeldin A，ATPase 抑制剂）和莫能菌素（Monensin）。PMA 刺激细胞激活 Protein kinase C（PKC），可以引起下游众多蛋白激酶的磷酸化，形成级联反应，引起细胞活化。Ionomycin 是一种来源于 Streptomyces conglobatus 的聚醚类窄谱抗生素，是一种有效的 Ca2+载体，高度选择性结合 Ca2+（也能转运其他离子，但亲和力相对较低）。Ionomycin 通过直接刺激钙池调控钙内流，提高胞内 Ca2+水平。PMA 常与 Ionomycin 联用，刺激免疫细胞活化。同时加入蛋白转运抑制剂Brefeldin A 和 Monensin 阻止高尔基体将新分泌蛋白运输到细胞外，以增强细胞内细胞因子的检测。
