Multicolor flow cytometry panel to identify human T cells, B cells,and NK cells. PBMCs were stained with anti-human CD45 ABflo® 488, CD3 Percp-Cy5.5, CD4 ABflo® 594, CD8a APC, CD19 PE, CD56 APC-Cy7. All antibodies are validated for cytometry. CD4 T cells are defined as CD3+ CD4+; CD8 T cells are defined as CD3+ CD8a+; B cells are CD3- CD19+; NK cells are CD3- CD56+; NKT cells are CD3+ CD56+.
The 6-fluorescence Human TBNK panel has been designed for reliable identification of human T, B, and NK lymphocyte populations in human blood or tissue samples.
ABflo® multi-fluorescence panel RK40002 covers the core CD markers of CD45, CD3, CD19, CD56, CD4, and CD8a. Our plots showed clear subgroups of CD3+ T cells, CD4+ Th cells, CD8+ CTL cells, CD19+ B cells, CD56bright NK cells, CD56dim NK cells, CD19+ CD56+ NK cell subsets, and CD3+ CD56+ NK T cells (cite figures).

Note:
This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1×10^6 cells in a 100 µl experimental sample (a test).
CD45: The leucocyte common antigen, protein tyrosine phosphatase receptor type C (PTPRC), is a transmembrane glycoprotein, expressed on almost all haematopoietic cells except for mature erythrocytes. CD45 is a central player of immune cell activation.
CD3: Is a complex composed of 4 distinct subunits, Delta (CD3ƍ), Epsilon (CD3ε), Gamma (CD3γ), and Zeta (CD3ζ). It Is a surface structure associated with the T-cell receptor (TCR) to form a complex involved in antigen recognition and signal transduction. CD3 is a pan-T-cell marker and is expressed by thymocytes and all mature T cells of the peripheral blood and lymphoid tissues.
CD19: Is a 95 kDa transmembrane glycoprotein belonging to the immunoglobulin (Ig) superfamily. CD19 is one of the most reliable surface biomarker for B cells. It is expressed from pre-B cells until the terminal differentiation to plasma cells.
CD56: Is also known as the neural cell adhesion molecule (NCAM), and a member of the immunoglobulin superfamily. CD56 is a fundamental marker in the determination of human natural killer (NK) cell subsets.
CD4: Is a 55 kDa transmembrane glycoprotein expressed on helper T lymphocytes. CD4⁺ T cells are crucial in achieving a regulated effective immune response to pathogens.
CD8a: Is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. CD8⁺ T-cells are important effector cells in specific cell-mediated antiviral immunity.
首先，一般抗体不推荐客户回收利用，抗体使用之后缓冲体系已经发生改变，不同客户在回收抗体的保存条件上也会有差异，所以抗体回收使用效果无法保证。另外，ABclonal公司也做过一批抗体回收验证测试，测试结果显示不同抗体可回收次数不同，一般效价越高的抗体，可重复使用的次数越多，客户可根据实验情况来确定。
注：我们将孵育完毕后剩余的抗体回收到离心管中置于4℃保存，效价高的抗体可至少保存1周，至少重复利用3次。

ABclonal抗体成分在平时工作当中，常会有客户咨询我们的抗体用的什么buffer进行保存，一般来说，我们的buffer的成分是：PBS含0.03%的proclin300、0.05%牛血清白蛋白、50%甘油；也有一些是PBS含0.03%的proclin300，50%甘油。防腐剂 Proclin 300活性成分主要是2-甲基-4-异噻唑啉-3-酮（MCI）和5-氯-2-甲基-4-异噻唑啉-3-酮（CMCI）。ProClin生物灭活剂能够迅速穿透细胞膜，抑制对细胞呼吸至关重要的特定酶，因此一接触微生物有机体就会立即抑制细胞活性。ProClin的多个特定毒性位点可以防止微生物产生高水平的耐药性。
